Deuterated Polyunsaturated Fatty Acids Reduce Oxidative Stress and Extend the Lifespan of C. elegans

Chemically reinforced essential fatty acids promise to flight numerous age-related diseases including Alzheimer’s, Friedreich’s ataxia and other neurological conditions. The reinforcement is achieved by substituting the atoms of hydrogen at the bis-allylic methylene of these essential fatty acids with the isotope deuterium. This substitution leads to stronger chemical bonds and, hence, the prevention of substantial chemical damage.

RT001 to treat neurodegeneration—Case Study: Improvement in a single patient with Late Onset Tay-Sachs Disease (LOTS)

Objective: We report on the use of RT001 in a single patient with LOTS.

Conclusions:

1. D-LA is elongated to D-AA; both achieve significant plasma levels and RBC membrane incorporation within 1 month of administration.

2. Early efficacy signs of inhibition of disease progression and some regression were seen.

3. RT001 was well tolerated with no toxicities reported.

Threshold Protective Effect of Deuterated Polyunsaturated Fatty Acids on Peroxidation of Lipid Bilayers

Autoxidation of polyunsaturated fatty acids (PUFAs) damages lipid membranes and generates numerous toxic by‐products implicated in neurodegeneration, aging and other pathologies. Abstraction of bis‐allylic hydrogen atoms is the rate‐limiting step of PUFA autoxidation, which is inhibited by replacing bis‐allylic hydrogens with deuterium atoms (D‐PUFAs). In cells, the presence of a relatively small fraction of D‐PUFAs among natural PUFAs is sufficient to effectively inhibit lipid peroxidation. Here, we investigate the effect of various D‐PUFAs on the stability of liposomes under oxidative stress conditions.

Parents of Leeds toddler with rare condition thank public as USA drug trial continues

The toddler was one of only 15 children worldwide picked for the trial and since launching a fundraising effort, parents Christine Hamshere and Steven Lightfoot have seen people donate more than £22,000 towards treatment with the company Retrotope’s drug in New Jersey.

Zoe is taking two capsules of the drug – a type of fatty acid – each day, which will increase to three doses after 30 days.

The family, who attended the Goryeb Children’s Hospital in Morristown, New Jersey, will need to go to America again in six months for another half-a-year’s supply of the drug, and then again in January 2020.

Retrotope presented positive findings from a compassionate use study of the company’s lead candidate, RT001, in a patient with Infantile Neuroaxonal Dystrophy (INAD), at the 70th AAN Annual Meeting. The study showed that administration of RT001 to a ~2.5-year-old patient appears to have arrested disease progression and led to re-acquisition of lost developmental milestones.
